Meth. 32, 11-29 (1980) It was found that carbonyl iron treatment of whole blood followed by density gradient sedimentation of the iron containing cells resulted in the best recovery of cells but generally failed to efficiently deplete esterase-positive cells (MjO). Sephadex G-10 columns consistently depleted M(t) to low levels and provided cell yields superior to nylon and rayon wool columns. In contrast to nylon wool columns, using Sephadex G-10 columns it was possible to deplete Mp without altering the proportions of T- and B-lymphocytes. Sephadex G-10 columns of 5-6 ml bed volume loaded with 3-5 X 10^ cells proved optimum for maximum recovery of lymphocytes and depletion of M^. - (8 tab., 1 fig., 35 ref.) H. D. Meth. 32, 31-39 (1980) Using chromium-labelled mononuclear cells the movements of lymphocytes in Isopaque-Ficoll (I-F) gradients were analysed Even small percentages of ox or autologous human erythrocytes mixed with the lymphocytes caused a significant loss of lymphoid cells and this loss was augmented when higher numbers of red cells were added. Such losses could be prevented using I-F gradients of higher densities, but then neutrophils and erythrocytes were found to contaminate the mononuclear suspensions recovered. Purified B and T lymphocytes were found to move identically on the gradients, but when human erythrocytes were added to the lymphocyte suspensions to be separated, a small but significant preferential loss of T cells was observed - (3 tab., 3 fig., 15 ref.) This absorption procedure was more rapid than ion exchange chromatography on DEAE cellulose or molecular sieving on Sephadex G-200 and gave a better yield of functionally purer antibody This absorption method gave antiserum suitable for the preparation of either IgM or IgG sensitized sheep erythrocytes and should be of value for the large-scale preparation of indicator cells required for the study of complement action and for detection of specific receptors on cell surfaces. - (1 tab., 3 fig , 12 ref.)
