The technique employs a series of small chambers formed on a microscope slide by the application of a plastic film in which holes have been punched. The procedure uses 5xl04 cells and 25pl of serum per chamber, and each test can be completed in 2 h. A permanent preparation results which can be examined as and when convenient. Meth. 20. 109 - 115 (1978) The percentage of non-immune rosettes varies according to the method of separation of lymphocytes. Buffy coat on Dextran-70 and Gravity Sedimentation method give the best rosette yields as compared with lymphocytes separated by centrifugation of heparinized blood (unsupported cen-trifugation) (P<0.001) or on Hypaque-Ficoll or Diaginol-Ficoll gradients (supported centrifugation) (P<0.05). The method entails reacting Ig-capped lymphoid cells with allo-antisera, and then assessing the uptake of alloantibodies by resetting the lymphocytes with SRBC coated with sheep IgG specific for mouse Ig. This resetting procedure was found to be generally more sensitive than the conventional dye exclusion microcytotoxicity test for detecting the binding of alloantibodies to lymphocytes.
